How Humanized Mouse Models Provide Closer Replication of Human Cancer Pathology

metastasis – providing valuable insights into oncogenesis mechanisms, validation of novel therapeutic targets, and as a predictive platform to test chemopreventives and treatments. Developing humanized mouse models that replicate both the human disease pathology and an intact (human) immune system are a top priority to facilitate the challenge of translating biologics and enable evaluation of concomitant therapies.
Mouse models have been developed which offer robust methods to study both potential treatments and preventives as well as the pathogenic and metastatic properties of Colorectal Cancer (CRC).Conditional knockout mouse models are useful for mitigating undesired symptomology, such as the co-development of lymphoma with intestinal neoplasia in MMR-disrupted mouse models – addressed by a mouse model similar to HNPCC wherein Msh2 is knocked down in villin-expressing tissues and MMR activity is preserved in the rest of the body1. Familial adenomatous polyposis (FAP) also involves a germline mutation in one Apc allele – Apc mutant mice (e.g. FabplCre; Apc15lox/+) are now capable of producing adenomas that are similar to the adenogenesis observed in FAP patients. Additionally, studies have shown that the location of the mutation within the Apc gene has an influence on adenoma multiplicity.

To all researchers:

During the AAALAC accreditation visit at the Pre- Clinical Research Authority held on February 23^rd, and February 24^th one of the two major issues that concerned the AAALAC delegates was the use of expired drugs.

This issue was addressed as mandatory for immediate correctional action to be taken in order to attain AAALAC accreditation.

Expired drugs and unmarked beyond-use-date open solutions, were found in several locations including researchers’ lockers, cabinets & laboratories.

Locked researchers cabinets used for drug storage within the PCRA locations emphasize the need of the PCRA staff to gain access for monitoring purposes.

In order to avoid such future concerns, all researchers and PCRA staff are requested to act in accordance with the following guidelines:

Drug expiration and expiration labeling:

Drugs and solutions for injections or for drug dilution should be used before the expiration date marked on the compound.

A single-dose or single-use vial is a vial of liquid medication intended for parenteral administration (injection or infusion) that is meant for use in a single patient for a single case, procedure, injection. Single-dose or single-use vials are labeled as such by the manufacturer and typically lack an antimicrobial preservative.

Vials that are labeled as single-dose or single-use should be used only for a single procedure/injection/ day and should be discarded when opened at the end of the day.

A multi-dose vial is a vial of liquid medication intended for parenteral administration (injection or infusion) that contains more than one dose of medication. Multi-dose vials are labeled as such by the manufacturer and typically contain an antimicrobial preservative to help prevent the growth of bacteria.

 The preservative has no effect on viruses and does not protect against contamination when personnel fail to follow sterile procedures of withdrawal from the vial.

 Medication vials should always be discarded whenever sterility is compromised or cannot be confirmed.

If a multi-dose vial has been opened or accessed (e.g., needle-punctured) the vial should be dated and discarded within 28 days unless the manufacturer specifies a different (shorter or longer) date for that opened vial.

All vials while in use should be stored according to manufacturer’s instructions

If a multi-dose vial has not been opened or accessed (e.g., needle-punctured), it should be discarded according to the manufacturer’s expiration date.

The manufacturer’s expiration date refers to the date after which an unopened multi-dose vial should not be used.

The beyond-use-date refers to the date after which an opened multi-dose vial should not be used. The beyond-use-date should never exceed the manufacturer’s original expiration date.  The beyond-use-date should be noted on the drug when first opened.

Solutions for injections or  for drug dilution if not  opened or accessed (e.g., needle-punctured) should be discarded according to the manufacturer’s expiration date. If opened or accessed (e.g., needle-punctured) should be  discarded within 7 days.   The beyond-use-date should be noted on the drug when first opened.

Per os. /Syrups, Ointments and creams if not opened should be discarded according to the manufacturer’s expiration date. If opened, should be discarded within 7 days unless the manufacturer specifies a different (shorter or longer) date. The beyond-use-date should be noted on the drug when first opened.

Drugs storage and expiration monitoring

Drugs (not including controlled substances) will be stored in locked cabinets.

Drugs will be stored in their original packages and in accordance with the manufacturer storage guidelines.

PI’s are responsible for monthly checks of the storage conditions and drugs expiration dates.

Expired drugs (not including, anesthetic drugs and narcotics) will be documented (Form f-044-2) and discarded as chemical waste.

Researcher’s drug cabinets within the PCRA facility should be available for inspection by the facility veterinarian at any time.

Drugs kept within researchers labs outside the PCRA facility should be kept and checked in the same manner. The facility veterinarian or IACUC committee will carry out additional inspections on their visits.
